Melatonin and Sleep: a Review

A total of 199 human studies (196 different publications) on the relationship between melatonin and sleep, were found through a search on Pubmed, the Internet medical Medline databank: 58 observational and 141 treatment studies.  71% of the observational studies showed a beneficial positive association between the serum or urinary level of melatonin or of one of its metabolites and sleep.  98 of the treatment studies were placebo-controlled studies with 82 (84 %) of them showing significant beneficial results of melatonin supplementation on sleep.  Most frequent beneficial effects were a reduction of sleep onset, an efficient muscle relaxation, better resynchronisation of advanced or delayed sleep phase syndromes, and restorations of circadian temperature and sleep-wake cycles.   Slow wave sleep and REM stages were rarely significantly improved.
